Het plot
Een door mensen ontwikkeld virus met de naam "KV" roeit in 2009 de gehele bevolking van New York uit, waarbij de wetenschapper Robert Neville (Will Smith) de enige menselijke overlevende in de stad, en misschien wel in de wereld is. Neville woont drie jaar lang alleen, het is inmiddels 2012 en hij probeert nu contactte vinden met andere mogelijke overlevenden. Hij wordt gade geslagen door een soort mutanten. Neville was de indirecte oorzaak van het uitsterven van de mens: zijn researchprogramma droeg bij de creatie van het virus, nu doet hij er alles aan om er een genezing voor te vinden. Hij is immuun, en beseft dat 2% van de bevolking dat ook moet zijn. Maar een groot deel van hen is ook omgekomen door toedoen van de mutanten die tevens kannibalen werden. Neville weet dat voor de overlevenden iedere seconde telt.
